Achieving Consistency in Biologics Production through Precise Cell Culture and Fermentation

During upstream processing production (USP), the fermentation regime (batch, fed-batch, continuous, or perfusion) and process conditions determined and optimized in the pilot bioreactors are transferred and used for biologics production.

The cells are grown to a volume of several thousand liters and produce several kilograms of the desired product.

Precise monitoring and control of the Critical Process Parameters (CPPs) (temperature, agitation, pH, dissolved oxygen (DO), CO2, turbidity, oxidation reduction potential (ORP)), and bioreactor content weight are required to ensure process optimality and consistency to maximize yield and productivity rates.

Upstream Processing Steps

Bioprocessing steps, from the cryogenic vials to the main culture:

Thawing of the cryogenic vial.
Shake flask cultures, from 0.5L to 1L.
Seed cultures, from 10L to 200L.
Main culture, 1000L.

Cell culture media serve as an environment where the cells can expand, and it provides nutrients for the cells to grow and express the biomolecules of interest.

Cell culture and fermentation media must contain precise quantities of nutrients and be sterile, and CPPs—such as pH, dissolved oxygen (DO), and culture yield weight—are carefully monitored in situ and real-time to enable reproducible and reliable results during the bioprocess and downstream processing.

The cells are the starting material of the biologics biomanufacturing process.

To build a working cell bank or start a seed train, scientists rely on accurate liquid handling solutions for the passages required before cryopreservation or transfer into the expansion seed train.

The expansion seed train is used to progressively increase cell density and reach the target number of cells required for the production bioreactor.

Each step of the expansion process and the main culture require precise temperature, agitation, pH, DO, dCO2, turbidity, ORP monitoring,weight measurement and control to optimize the operation and ensure optimal production conditions.

At the end of the production process, the broth, the product, and the cells are collected to be separated and purified during downstream processing
